Iambic, an AI-focused drug discovery company with multiple big-pharma partnerships, said its planned IPO will help fund an internal oncology pipeline led by IAM1363, a Phase I/Ib HER2 inhibitor that has shown anti-tumour activity in heavily pretreated patients with HER2-altered solid tumours. The company also said it plans a registrational trial for IAM1363 next year and IND filings in the fourth quarter for two additional preclinical cancer programmes.
Why It Matters To Oncology
Iambic is trying to convert AI-enabled discovery revenue into internally owned oncology assets, a closely watched model for clinicians and drug developers.
Its lead candidate, IAM1363, is an oral, brain-penetrant small-molecule HER2 inhibitor in Phase I/Ib testing for HER2-altered solid tumours.
The company has reported anti-tumour activity in heavily pretreated patients across tumour types including non-small-cell lung, breast, gastric, renal, ovarian and other HER2-driven cancers.
IAM217, a brain-penetrant allosteric KIF18A inhibitor, is being developed for ovarian cancer, triple-negative breast cancer and other solid tumours, while IAM-C1 targets HR-positive, HER2-negative breast cancer.
The Financials
Iambic disclosed its IPO plans shortly after announcing a new discovery collaboration with AbbVie.
Earlier this year, it also signed deals with Takeda and Bayer; the Takeda agreement is worth up to $1.7 billion and includes oncology, gastrointestinal and inflammatory diseases.
The company said partnership income has supported operations so far, but proceeds from a public offering are expected to help advance its own pipeline.
Iambic added that some IPO proceeds could also be used for acquisitions or investments in products, services or technologies.
What They're Saying
Iambic says its Enchant v3 platform can integrate assay data, molecular and protein structures, multi-omics and pharmacokinetics to predict candidate properties and compare drug-design strategies.
In its securities filing, the company said IAM1363 is the only HER2 tyrosine kinase inhibitor that binds the inactive conformation of the HER2 kinase domain.
That mechanism, Iambic argues, may support selectivity for HER2 over wild-type EGFR and broad activity across HER2 variants with activating mutations.
NVIDIA, which is also collaborating with Iambic to optimise AI models, holds a stake of more than 5%, according to the filing.
What's Next
Iambic plans to begin a registrational trial for IAM1363 in 2027.
The company is also studying IAM1363 with Jazz Pharmaceuticals' bispecific HER2 antibody zanidatamab in heavily pretreated HER2-positive breast cancer.
IND filings for IAM217 and IAM-C1 are planned in the fourth quarter.
For oncology clinicians, the near-term watch points are durability and breadth of HER2 activity, CNS relevance, combination data and whether AI-enabled discovery can translate into registrational success.
